Analysis of interleukin-8 -251A/T polymorphism and IL-8 mRNA expression in sporadic colon cancer

Colon cancer tumorigenesis is a multistep process of mutation accumulation in a number of oncogenes and tumor-suppressor genes. Crosstalk between normal and tumor cells is recognized to influence various stages of this process. Cytokines that are produced in the tumor microenvironment have an important role in cancer pathogenesis. Cytokines are often encoded by polymorphic genes. Single nucleotide polymorphisms (SNPs) in the promoter regions of these genes are associated with differential levels of cytokine expression. Polymorphism in genes regulating the immune response and cell growth may result in increased susceptibility to and/or poorer prognosis in certain cancers. Interleukin-8 (IL-8) might influence colon cancer tumorigenesis through affecting the efficiency of the anti-tumor immune response and/or angiogenesis. The aim of this study was to examine IL-8 mRNA expression in sporadic colon cancer and corresponding normal mucous tissue, as well as to examine the frequency of IL-8-251A/T polymorphism in sporadic colon cancer and its possible influence on IL-8 mRNA expression. Real-time TaqMan&reg ; RT-PCR was used to determine the IL-8 mRNA expression in 60 pairs of colon cancer tumors and corresponding normal tissue. A statistically significant difference in IL-8 mRNA expression was observed. IL-8 mRNA expression was higher in tumor tissue as compared to corresponding normal mucous tissue. A total of 160 colon cancer patients and 160 unrelated cancer-free controls were genotyped for IL-8-251A/T SNP using real-time PCR TaqMan&reg ; SNP genotyping assay. No significant association was found between sporadic colon cancer and IL-8-251A/T genotypes. No significant association was found between different IL-8-251A/T genotypes and IL-8 mRNA expression, as well. The results of our study indicate that IL-8 mRNA expression was higher in tumor as compared to normal tissue. However, IL-8-251A/T SNP genotypes were not associated with differential IL-8 mRNA expression and IL-8-251A/T did not influence sporadic colon cancer susceptibility, although a role of other IL-8 gene polymorphisms cannot be excluded.
