Journey from Brush-type to Polysaccharide CSPs
Our team is developing new chiral stationary phases (CSPs) based on thorough knowledge of mechanism of chiral recognition, most distinguishing property of brush-type CSPs. Conceptually, we recognize linear “ end on” and linear “ bifurcated” , tweezer-type and dendrimer-type CSPs, and for any particular group we expect specific properties in molecular recognition process. It is interesting to note that respective chiral selectors belong to C1, C2 and C3 symmetry group. Our scientific break-trough came with discovery of a family of tweezer-type structures, followed by effective linear and tethered structures, which proved highly effective for separation of broad spectrum of racemates. Examples for enantioseparation of alpha-aryloxypropionic acids and aminoalcohols will be presented. Detailed study of enantiorecognition mechanism in separation of chiral dihydropyrimidones (DHPM), in particular difference in chromatographic behaviour of monastrol and its oxo-analoges, with new CSPs will be presented. Determination of chromatographic parameters for separation of racemic BINAPO has enabled its SMB separation on specially designed CSP, and opened new commercial approach to both enantiomers of BINAP. Attracted by versatility in enantioseparation and commercial success of polysaccharide based CSPs we entered study of some inventive solutions in their preparation and their application. Examples for enantioseparation of atenolol, bambuterol and amlodipine will be presented.
