Molecular cytogenetic confirmation of hybrids between Iris variegata and Iris illyrica in Dinaric Alps
In the region of Littoral Dinaric Alps (Mt Velebit, Croatia) where the two Iris species (I. variegata L. and I. illyrica Tomm.) grow in sympatry, many morphologically intermediate individuals were observed. The aim of this study was to examine the possible hybrid nature of these individuals by using the molecular cytogenetic techniques. The karyotype characterisation of studied species and hybrid like individuals was performed by using fluorochrome banding (Chromomycin A3, and DAPI - 4', 6-diamidino-2- phenylindole) and double Fluorescence In Situ Hybridization (FISH) with 18S-5.8-26S and 5S rDNA probes. In this way, the hybrid origin of these individuals was confirmed by molecular cytogenetic investigations. The results of fluorochrome banding and FISH experiment for I. variegata, I. illyrica and their hybrid were reported here for the first time. Differences occurred between the parental species in the number and position of chromomycin bands, as well as 18S-5.8S-26S and 5S rDNA loci. Thus, the hybrid karyotype pattern presents the intermediary characters between two parental species. Furthermore, some hybrid individuals showed different karyotype patterns which can indicate their possible origin from back-cross with one of parents.
