H-ras intron 1 hexanucleotide polymorphism in sporadic colon cancer
Colon cancer arises from multiple changes at the cellular level. Mutations in number of genes occure in its carcinogenesis. One of the genes that are considered to be involved in the development of sporadic colon cancer is H-ras. The aim of this study was to investigate one of the polymorphisms inside H-ras gene – hexanucleotide polymorphism in the first intron. Two hundred healthy volunteers were genotyped in order to determine the distribution of alleles and genotypes at this polymorphic locus in Croatian population. To determine if it is connected with susceptibility to colon cancer in our population we analyzed two hundred colon cancer patients and compared the obtained results. We also compared the results between genders to find if the distributions were gender-related. In our study LOH of H-ras gene using hexanucleotide polymorphism in sporadic colon cancer was also investigated. We noticed statistically significant difference in genotype distribution at hexanucleotide locus between healthy population and colon cancer patients (p=0.0128735) but not in allelic distribution. No differences were found between genders. LOH was found in 10% of tumour samples.
