Genetic Charcterization of Sweet Chestnut Cultivars in Croatia
The sweet chestnut orchards for the production of high quality nuts in Croatia have been established in Opatija and Lovran region, from different cultivars, but there is no data on their origin and there is a complete absence of any description in terms of name (all of them are called 'Marrone'), morphological, phenological or genetic characteristics. There is a great need for the improvement and recovery of old and abandoned orchards. In order to preserve the endangered biodiversity of the cultivated chestnut germplasm, it is necessary to identify, name and describe existing cultivars. The main objective was to characterize the sweet chestnut cultivars using microsatellite markers. The leaf samples have been collected from 96 old grafted trees. DNAs were isolated from fresh leaf tissue and microsatellite analysis was performed using ten microsatellites primers. The results showed a high degree of genetic variability.
