The role of some bioactive dietary compounds in modulation of colorectal cancer

Colorectal cancer is considered to be a diet- related type of cancer and is the third most common type of cancer in the world. In this abstract, we present the relevant dietary patterns involved in neoplastic cell growth as well as protective components in colon cancer development. Dietary polyamines (putrescine, cadaverine, spermidine and spermine) are involved in many physiological functions ; however, they can be also involved in neoplastic cell growth. Thus, a polyamine-low diet has been proposed as part of the nutritional therapy for cancer patients. Generally, polyamine levels can be high in decomposed fish and fermented food products (fish, shellfish, meat, vegetables). Furthermore, high dietary biogenic amines levels (tyramine, putrescine, cadaverine) can act as precursors of N-nitroso compounds (nitrosamines), which are carcinogens [1]. On the other hand, some bioactive dietary components such as omega-3 polyunsaturated fatty acids, resveratrol, curcumin, quercetin, vitamin D and dietary fiber have preventive effects in colon cancer development. These components can modulate miRNA expression modifying several important pathways including carcinogenesis (from initiation to tumor promotion), cell proliferation, angiogenesis, apoptosis and metastasis [2]. Thus, dietary based miRNA manipulation can present potential benefit for prevention and treatment of carcinogenesis. Furthermore, beta-caryophyllene (BCP), a bicyclic sesquiterpene, is a phytocannabinoid with strong affinity to cannabinoid receptor CB2 and anticancer- associated effects, which is particularly valuable in oncology. BCP and BCPO (BCP oxidation derivate) are common bioactive components of essential oils in a large number of spices, food plants (oregano, black paper, basil, cloves, cinnamon, rosemary, lavender, and cannabis) as well as Cannabis. The molecular mechanism of BCP anticancer activity includes activation of the CB2 receptor which triggers ceramide-ERK signaling, activation of the p38MAPK pathway and the activation of caspases which promote apoptosis. Although BCPO does not display any affinity toward cannabinoid receptors, its anticancer activities include activation of the MAPK pathway, and inhibition of PI3K/AKT/mTOR/S6K1 and STAT3 signaling in cancer cells [3]. Additionally, BCPO enhances the anticancer effectiveness of chemotherapeutic drugs by increasing their cellular accumulation. Thus, the potential therapeutic utility of BCP/BCPO application in combination with classical anticancer drugs deserves further investigation.
