Identification of Fusarium species isolated from rotten apple fruit after storage

Fusarium species are primarly known as the causal agents of cereal and grass diseases, but they also occurr on a number of vegetable, ornamental or fruit species. It is known that Fusarium spp. can cause rot of apples and other fruits during and after storage. Fusarium rot may be caused by several species of genus Fusarium. Since Fusarium taxonomy is very complex and has been constantly revised and updated over the last years, identification of Fusarium species today is based on combined morphological characteristics, biological and molecular data. Totally 32 Fusarium isolates were recovered from rotten apple fruit of cultivars Golden Delicious, Jonagold, Idared and Pink Lady, stored in Ultra Low Oxygen (ULO) conditions. Observed symptoms of Fusarium rot were similar on all four cultivars, with soft circular brown necrosis of different extent, with or without visible sporulation. Species were identified according to the morphology on potato-dextrose agar (PDA) and carnation leaf agar (CLA). 21 isolates were identified as Fusarium avenaceum and were confirmed with polymerase chain reaction (PCR) using specific primer pair FA-ITSF and FA-ITSR. F. pseudograminearum, F. semitectum and F. crookwellense were identified based on morphological characteristics. It was concluded that F. avenaceum was the dominant species causing Fusarium rot of apple fruit after storage. Pathogenicity tests were conducted, and most of the Fusarium isolates caused typical symptoms of Fusarium rot when inoculated on wounded apple fruits. On the other hand, only a few isolates caused necrosis when inoculated on unwounded fruits, and such results suggested that Fusarium species on stored apple fruits are typical wound parasites.
